ASO
Antisense oligonucleotides (ASOs) are single-stranded oligonucleotides molecules with 15-25 nucleotides in length. After entering the cell, ASOs are bound to their respective complementary target mRNAs under the action of ribonuclease H1 (Rnase H1) upon the principle of complementary base pairing, which inhibits the expression of target genes.
Phosphate backbone modification: Oxygen atoms in ASO phosphate groups are replaced by sulfur atoms to form a phosphorothioate (PS) backbone.
Sugar ring 2' position modification: the most commonly used types of ASO modification are 2′-O-methylation (2'-O-Me) and 2′-O-methoxyethyl (2'-MOE).
